The protein expression of prostate and ovarian tumor cells are studied to search for possible markers of cancer. Fresh frozen tumor cells are obtained and prepared for study by the pathology groups at the University of Michigan Medical Center. The tumors are lysed and separated utilizing a 2-D liquid separation, which is capable of separating out practically 3000 protein bands. This strategy involves separation based upon pI in the 1st dimension making use of chromatofocusing and by hydrophobicity in the second dimension making use of nonporous silica (NPS) RP-HPLC.
The liquid eluent from the second dimension is then split and 1/three is directed to an electrospray time-of-flight mass spectrometer (ESI-TOF MS) for mass analysis, although the remaining 2/three is collected by a fraction collector for MALDI-TOF MS analysis. The result is a mass map as a function of pI analogous to two-D gel electrophoresis. The ESI-TOF MS gives an accurate MW value, which can be used with MALDI-TOF MS peptide mapping and MS/MS for accurate identification of proteins in these human cancer cells. In addition, the MW value provides a implies to decide regardless of whether a protein is modified which includes the presence of truncations, deletions, sequence adjustments and splice variants.
Using this strategy significant numbers of tumor samples can be mapped and every form of cancer can be classified according to its protein profile. In addition, marker proteins have been identified which can be associated with distinct grades and stages of cancer progression. The method supplies a signifies for pathologists to identify the grade of the tumor, i.e. whether it is a lot more benign or malignant, and consequently establish a distinct therapy for that tumor.

About Dr. David M. Lubman:
David M. Lubman received his Ph.D. in physical chemistry from Stanford University in 1979 under the direction of Professor Richard N. Zare. He was a fellow at the Weizmann Institute of Science and arrived at the University of Michigan in 1983 as a member of the Department of Chemistry.
He was promoted to Associate Professor in 1987 and Full Professor in 1991. Among a number of awards in the field, he has won an Alfred P. Sloan Foundation Award, the Meggars Award of the Society of Applied Spectroscopy, an R & D 100 Award and was recently elected as a Fellow of the American Association for the Advancement of Science. He has also won internal awards inside the University of Michigan which includes the LS&A Excellence in Analysis Award, LS&A Excellence in Teaching Award and the Faculty Recognition Award.
His present investigation interests involve developing new proteomic techniques for looking for protein markers of cancer. These include two-D liquid mapping of tumor cells and serum and also the development of protein and aptamer microarray technologies. A variety of collaborations with clinical researchers consist of studies in prostate, ovarian and breast cancer.
Dr. Lubman presently serves on the Editorial boards of J. Chromatography, Existing Analytical Chemistry, Anal Chim Acta, and Mass Spectrometry Evaluations. He is a member of American Association for the Advancement of Science, American Chemical Society and the American Society for Mass Spectrometry.
